牛排菇SB65菌株液体培养基和培养条件的优化
Optimization of fermentation liquid medium and culture conditions of portabella mushroom(Agaricus brunnescens) SB65 strain

摘要: 通过单因素实验和均匀设计对牛排菇SB65菌株的液体菌种培养基和培养条件进行了优化.优化后的培养基配方为(w/v):蔗糖5.5%,酵母浸粉1%,MgSO4·7H2O 0.05%,KH2PO4 1%,NaCl 0.7147%,CaCl2·2H2O 1%;液体培养条件为:初始pH=7.2,培养温度28℃,培养时间12 d,摇床转速160 r/min,装液量50 mL/250 mL三角瓶,接种量8%(v/v).在优化的培养基和培养条件下进行培养,菌丝体的生物量为4.579 g/L.Abstract: The liquid medium and culture conditions of portabella mushroom(Agaricus brunnescens) SB65 strain were optimized by single-factor experiments and uniform design.The optimized medium were as follows(w/v):sucrose 5.5%,yeast extract 1%,MgSO4·7H2O 0.05%,KH2PO4 1%,NaCl 0.714 7%,CaCl2·2H2O 1% and the optimized culture conditions were as follows:initial pH=7.2,culture temperature 28℃,culture period 12 d,rotation rate 160 r/min,medium volume 50 mL per 250 mL flask and seed volume 8% (v/v).Under the optimized medium and culture conditions,the biomass of tested strain was 4.579 g/L.
