6-羟基烟碱与烟碱的安全性比较研究

摘要: 通过毒理学试验比较6-羟基烟碱与烟碱的安全性:对SPF级雄性小白鼠(体重30~35 g)进行急性毒性试验,分别进行35 d连续灌胃每只1.00 mg,0.75 mg,0.50 mg,0.25 mg的烟碱和6-羟基烟碱试验(分别相当于每d抽吸20支烟的烟民烟碱接受量的8倍,6倍,4倍,2倍);计算各组心肝脾肺肾的脏器系数,检测血液中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、甘油三酯(TG)3项生化指标,并对小鼠活体解剖取肝、肺、肾组织作病理学检查.结果表明,6-羟基烟碱相比烟碱的毒性明显降低(P < 0.05),低剂量的烟碱不会造成小鼠器官的病变.Abstract: he safety of 6-hydroxynicotine and nicotine was compared through toxicology test.The acute toxicity and accumulative test was carried out on SPF mice(each of 30~35 g).After 35 d intragastrically each of 1.00 mg,0.75 mg,0.5 mg,0.25 mg of nicotine and 6-hydroxynicotine(about receive the amount of a man pumping 20 cigarettes a day 8,6,4,2 times),the organ coefficient of heart,liver,spleen,lung and kidney were calculated and bloodindicators of ALT,AST,TG were determined,the pathological examination was also done using liver,lung and kidney tissue.The results showed that 6-hydroxynicotine compared to nicotine significantly reduced in toxicity(P < 0.05),low doses of nicotine does not cause organ lesions in mice.
