抗菌PLGA薄膜的初步研究

摘要: 以不含端羧基的聚乳酸-羟基乙酸共聚物(PLGA)薄膜为材料,分别采用氢氧化钠处理和紫外线处理使其加上端羧基,利用原子力显微镜对处理后的PLGA薄膜表面进行表征,并对其亲水性能进行检测.运用碳化二亚胺两步接枝法在含有端羧基的PLGA薄膜表面接枝活性蛋白杆菌肽,分别采用吖啶橙荧光染色法和MTT法来检测接枝前后PLGA薄膜的抗菌性能.结果表明:分别经氢氧化钠处理和紫外线处理后的PLGA薄膜表面化学结构均发生了明显变化;与空白薄膜对比,经氢氧化钠处理的PLGA薄膜的亲水性变大;与未接枝杆菌肽的PLGA薄膜进行对比,接枝后的PLGA薄膜具有一定的抗菌作用,且抗菌性随杆菌肽接枝浓度的增加而逐渐减弱.Abstract: Taking the thin film of PLGA without carboxyl end group as the material, sodium hydroxide and ultraviolet ray were adopted respectively to make it with carboxyl end group. Atomic force microscopy was used to characterized the thin film of PLGA after treatment, and its hydrophily was tested. Then grafting the active protein bacitracin through the carbodiimide sequential method, and the antibacterial performance of the thin film of PLGA before and after grafted was tested respectively by the methods of acridine orange fluorescence staining and MTT method. The results showed that the chemical structure of the thin film of PLGA dealt with sodium hydroxide and ultraviolet ray changed obviously. Compared with the blank thin film, the hydrophily of the thin film of PLGA after treatment became bigger. Compared with the thin film of PLGA without grafting, the thin film of PLGA after grating had the certain antibacterial performance, and the antibacterial performance gradually decreased as the grating concentration increased.
