烟叶中β-胡萝卜素高效降解菌株的筛选鉴定及发酵条件优化
Screening and identifying of β-carotene degrading strains from tobacco leaves and its optimization of fermentation conditions

摘要: 从新鲜的豫烟13烟叶中筛选能高效降解β-胡萝卜素的菌株,用GC-MS对其主要降解产物进行检测,利用形态学和16S rDNA进化树对菌株进行鉴定,通过单因素试验和正交试验对菌株的发酵条件进行优化。结果表明:分离到的YT-3菌株为霍氏肠杆菌亚种(Enterobacter hormanii subsp.),该菌株能高效降解β-胡萝卜素,其降解的主要产物为β-紫罗酮、二氢猕猴桃内酯、β-环柠檬醛等香味物质;YT-3菌株的最佳发酵条件为蔗糖质量浓度30 g/L、硝酸钠质量浓度4 g/L、酵母粉质量浓度3 g/L、初始pH值7.0,在该发酵条件下β-胡萝卜素降解率达到97.05%。Abstract: The strains that could degrade β-carotene with high efficiency were screened from fresh Yuyan 13 tobacco leaves, and the main degradation products were detected by GC-MS, the species of the strain were identified by morphology and 16S rDNA phylogenetic tree, the fermentation conditions of the strain were optimized by single factor test and orthogonal test. The results showed that YT-3 strain was Enterobacter hormanii subsp.which could degrade β-carotene with high efficiency. The main degradation products were β-ionone, dihydrokiwifruit lactone, β-cyclic citral (3.21%) and other flavor substances. The optimum fermentation conditions of YT-3 strain were as follows: NaNO3 concentration 4 g/L, sucrose concentration 30 g/L, yeast powder concentration 3 g/L, initial pH value 7.0. Under these fermentation conditions, the degradation rate of β-carotene reached 97.05%.
